Species reactivity
Dog; Due to limited knowledge and inability for testing each and every species, the reactivity of the antibody may extend to other species which are not listed hereby.
Description
This antibody needs to be stored at + 4°C in a fridge short term in a concentrated dilution. Freeze thaw will destroy a percentage in every cycle and should be avoided.
Properties
If you buy Antibodies supplied by MBS Monoclonals they should be stored frozen at - 24°C for long term storage and for short term at + 5°C.Canine or Canis Lupus is mostly Beagle used for drug research. MBS Monoclonals produces ELISA test kits and polyclonal antibodies.
Test
Mouse or mice from the Mus musculus species are used for production of mouse monoclonal antibodies or mabs and as research model for humans in your lab. Mouse are mature after 40 days for females and 55 days for males. The female mice are pregnant only 20 days and can give birth to 10 litters of 6-8 mice a year. Transgenic, knock-out, congenic and inbread strains are known for C57BL/6, A/J, BALB/c, SCID while the CD-1 is outbred as strain.
Storage and shipping
Store the antibody at +4 degrees Celsius for short-term storage and at -20 degrees Celsius for long-term.Storage in frost-free freezers is not recommended. the antibody should be stored undiluted. Repeated freeze - thaw cycles may denature the peptide chains of the antibody and therefore should be maximally avoided. If there is a precipitate in the vial we recommend you to briefly microcentrifugate it prior to use. Shelf Life: 18 months from date of dispatch.
Specificity and cross-reactivity
CD107b Mouse anti Dog CD107b (AC17) is a monoclonal antibody specific for canine CD107b, otherwise known as lysosome-associated membrane protein 2 or LAMP-2. Immunofluorescence staining of MDCK cells with Mouse anti Canine CD107b demonstrates staining patterns consistent with localization to lysozomes. This is supported by coincident staining of an exogenous lysozomal glycoprotein, avian LEP100 transfected into MDCK cells and detected using the anti LEP100 antibody clone CV24. Clone AC17 immunoprecipitates a protein of 95kDa in MDCK cells which, following Endo F digestion to remove N-linked oligosaccharides yields a core protein product of 40kDa, indicating the heavily glycosylated nature of CD107b. The molecular weight of canine CD107b is typical of many lysozome-associated membrane proteins. While most (97%) CD107b resides in the lysozomal environment in adherent MDCK cells in vitro, the early studies by indicate that a small percentage of total cellular CD107b as revealed by radioimmune assay with clone AC17, is found associated with the cell membrane. Lysosomes are membrane-bound organelles found within the cytoplasm of most cells, they contain hydrolytic enzymes and act as the major compartment for heterophagic and autophagic digestion. Members of the lysosomal-associated membrane protein family (LAMPS) are believed to play an important role in protecting the lysosomal membrane from protease degradation and are involved in lectin-mediated cell adhesion. CD107b has been shown to share high N-terminal amino acid sequence homology with human, mouse and rat CD107b; Since it is not possible to test each and every species our knowledge on the corss reactivity of the antibodies is limited. This particular antibody might cross react with speacies outside of the listed ones.
