Species reactivity
Mouse (Mus musculus); Due to limited knowledge and inability for testing each and every species, the reactivity of the antibody may extend to other species which are not listed hereby.
Description
This antibody needs to be stored at + 4°C in a fridge short term in a concentrated dilution. Freeze thaw will destroy a percentage in every cycle and should be avoided. This is against a protein of mus musculus directed antibody.
Properties
If you buy Antibodies supplied by MBS Monoclonals they should be stored frozen at - 24°C for long term storage and for short term at + 5°C.Armenian of Syrian hamster monoclonal IgGs are useful antibodies for double staining your IHC samples. Using mabs or monoclonal antibodies has a lot of advantages for Flow cytometry.
Storage and shipping
Store the antibody should be stored at -20 degrees Celsius. only. Storage in frost-free freezers is not recommended. the antibody should be stored undiluted. Repeated freeze - thaw cycles may denature the peptide chains of the antibody and therefore should be maximally avoided. If there is a precipitate in the vial we recommend you to briefly microcentrifugate it prior to use. Shelf Life: 18 months from date of dispatch.
Test
Mouse or mice from the Mus musculus species are used for production of mouse monoclonal antibodies or mabs and as research model for humans in your lab. Mouse are mature after 40 days for females and 55 days for males. The female mice are pregnant only 20 days and can give birth to 10 litters of 6-8 mice a year. Transgenic, knock-out, congenic and inbread strains are known for C57BL/6, A/J, BALB/c, SCID while the CD-1 is outbred as strain.
Specificity and cross-reactivity
CD339 This product recognises CD339, otherwise known as Jagged1, one of the five major ligands of the Notch signalling pathway, which is activated through the binding of specific ligands to the Notch receptors Notch 1-4. _x000D__x000D_The Notch signalling pathway is an evolutionarily conserved pathway in multi-cellular organisms, which is vital for cell-cell communication, important during fundamental developmental and physiological processes, including regulation of cell fate decisions during neuronal, cardiac and endocrine development, stem cell haematopoiesis, thymic T-cell development, and both tumour progression and suppression. _x000D__x000D_Ligation of Notch receptors by their specific ligands, Jagged1 (CD339), Jagged2, Delta like-1 (DLL1), DLL3 and DLL4, on physically adjacent signal receiving cells, induces proteolysis of the receptors by ADAM-family metalloproteases and gamma-secretase complex, within the transmembrane domain, releasing the Notch intracellular domain (NICD) to translocate to the nucleus. Subsequent signal transduction then occurs through either the CSL-NICD-Mastermind complex cascade (canonical pathway), or NF-kappaB-NICD and CSL-NICD-Deltex complex signalling cascades (non-canonical pathway). The canonical pathway inhibits the differentiation of stem cells or progenitor cells, whilst the non-canonical pathway promotes differentiation. _x000D__x000D_Jagged1 signalling is implicated in cell-fate decisions during haematopoiesis, as well as in both early and late stages of mammalian cardiovascular development, and is involved in the inhibition of myoblast differentiation. Studies have shown a significant increase in the expression of Jagged1 in metastatic prostate cancer, compared with localized prostate cancer or benign prostatic tissues, implicating Jagged1 as a biomarker to facilitate their differentiation. _x000D__x000D_In humans, mutations in the JAG1 gene are responsible for the autosomal dominant multi-system developmental disorder ; Since it is not possible to test each and every species our knowledge on the corss reactivity of the antibodies is limited. This particular antibody might cross react with speacies outside of the listed ones.
